Abstract
Amidst the genomic revolution in the characterization of the acute leukemias, immunophenotyping maintains a crucial position in diagnosis, determination of therapy, and monitoring of response to therapy. Lineage determination by multiparameter flow cytometry (MFC), the definition of surrogate marker profiles for genetic lesions which guides molecular analysis towards selective, thus limited, testing, MFC for universal MRD detection, monitoring of specific antigen expression targeted by monoclonal antibodies, and characterization of antigens which are associated with prognostically significant somatic mutations all provide valuable attributes to the comprehensive workup of the acute leukemia patient.
