Antimetabolites: A new perspective

Abstract

Antimetabolites remain as mainstays in our chemotherapeutic armamentarium. Recent approval of several nucleoside analogs and antifolates serve to highlight the progress made in the past 60 years but more importantly their pharmacology/biochemistry reminds us of the complexity of the pathways involved. Antimetabolites should be considered as specific targeting agents in the emerging arena of metabolomics and as tools to more completely understand cellular metabolism.
